If there is enough room behind the subwoofer, then yes, I mean line the back/bottom of the enclosure too.

so does lining the inside with fiberglass achieve the same benefits and more as the polyfill? you the polyfill fools the sub into thinking its in a bigger enclosure therefore it plays lower, does the glassing do the same?

Using fiberglass insulation (which is not the same as fiberglassing the inside of the enclosure because it is insulation, not fiberglass mat, so no resin is used) inside the enclosure helps damp any resonances, which results in smoothing out the frequency response. Using fiberglass insulation also aids in LFE (low frequency extension) because it also fools the subwoofer into thinking it's in a larger sized enclosure. Polyfill is still top dog as far as "adding" enclosure volume, but fiberglass damps the enclosure very well.
